Description: Das Projekt "Untersuchungen der Symbiose zwischen Rhizobium Meliloti und Alfalfa in sauren Boeden Argentiniens und Uruguays" wird vom Umweltbundesamt gefördert und von Universität Bielefeld, Lehrstuhl für Genetik durchgeführt. The main objective of this work is to further examine and improve the symbiotic ability of R meliloti strains from the central part of Argentina and Uruguay to be eventually used in inoculant production. For this propose specific aims have been devised: I) Development of a R meliloti strain collection with native rhizobia isolated from Argentina and Uruguay. Isolates from other regions of interest will be also studied. II) Development of appropriate methodologies to study competition for nodule occupancy. III) Application of these methods to the analysis of compentiveness among rhizobia from the strain collection: (Selection of strains with enhanced competitivity for nodule formation). IV) Analysis of N-fixing ability in plants nodulated with the strains selected as more competitive. (Selection of rhizobia with enhanced N-fixation) V) Phenotypic and genotypic characterization of strains that appear as symbiotically more active. Special attention will be paid to those pbenotypes related to strain competitiveness. Integration of result from points I to V. VI) Genetic manipulation of the strains to improve their symbiotic properties. Evaluation of results. Identification of genes involved in competition by analysis of cosmid gene bank from a highly competitive strain: Transfer of cosmids to recipient rhizobia with low profile of competitiveness and screening for strains with enhanced nodule occupancy. VII) Small scale plant inoculation experiments by using samples of soil. Emulation of field conditions. VIII) Technology transfer. During the whole work alfalfa varieties used in Argentina and Uruguay will be employed.
