Description: Das Projekt "Biologie und Diversitaet dunkler, septierter Baumwurzelendophyten (DSE) unter Verwendung molekular-genetischer Techniken" wird vom Umweltbundesamt gefördert und von Eidgenössische Technische Hochschule Zürich, Departement Wald- und Holzforschung, Professur für Forstschutz und Dendrologie durchgeführt. Dark septate root endophytes (DSE) are ubiquitous fungal tree root colonizers in temperate and boreal conifer forest ecosystems. Their biology and ecological significance is insufficiently known. In this project we propose to elucidate population structure and dynamics as well as mechanisms of reproduction of the two supposedly asexual DSE Phialocephala fortinii and 'Type 1'. The experimental programme will be multi-disciplinary in approach, utilizing classical mycological and plant pathological techniques as well as molecular genetic techniques. We intend to study the function of the conidia of P. fortinii and search for mating type genes in both P. fortinii and Type 1 (which is not known to sporulate). In addition, we envisage to study in detail the population structures of these fungi in a forest and the way they spread in soil and colonize roots. Norway spruce (Picea abies (L.) Karst.) will be used as model host plant.
