Es wird/wurde ausgeführt durch: Helmholtz-Zentrum für Umweltforschung GmbH - UFZ, Department Bodenökologie.Part 1: RootPatho-EM - Gene regulation and C and N fluxes during the endogenous rhythmic growth of oaks interacting with mycorrhizal as well as root pathogenic biotrophs. Trees produce photoassimilates and mobilize nutrients to support their own development but also associations with beneficial or detrimental biotrophic partners. Similar to major forest trees, oaks growth follows an endogenous rhythm with alternating flushes in root and shoot, which was shown to affect the formation of ectomycorrhizal symbioses between roots and fungi. The project part RootPatho-EM aims at enlightening the implication of this rhythm in interactions with the ectomycorrhizal fungus Piloderma croceum and the susceptibility against the root rot pathogen Phytophthora quercina. In controlled experiments with oak microcuttings inoculated with one or both microorganisms the plant resource allocation will be followed after labelling with 13C and 15N, while the plant gene regulation will be monitored with a broad range array. Additional experiments will investigate the impact of climatic fluctuations, tree neighbouring and interaction with actinobacteria. The project part RootPatho-EM also assumes the central function of producing the plant material and a mobile 13CO2/12CO2-labelling unit to analyse C allocation for the whole application, thus crucially contributs to its joined experimental platform (JEP) for rational cross comparison of gene regulation and resource allocation patterns in all considered biotrophic interactions. Part 2: RootCons- Effects of root and fungal feeding animal consumers on rhizosphere interactions and oak performance. The proposed project investigates the gene expression response of mycorrhizal and non-mycorrhizal oak microcuttings to major rhizosphere animals, root and fungal feeding invertebrates, represented by nematodes and Collembola, respectively. The gene expression response of the plants will be investigated using microarrays and qRT-PCR technology in parallel to carbon and nitrogen allocation patterns using 13CO2 and 15N pulse labelling. Two growth phases of oak microcuttings i.e., root and shoot flushes and their response to root and fungal feeders in the rhizosphere will be studied. In a further experiment the time dependent flux of carbon into rhizosphere compartments including roots, soil microorganisms and fungal feeders will be investigated using bulk 13C and compound specific 13C lipid analysis. Both separate and combined effects of nematodes and Collembola will be studied. In addition, interactions between root feeding nematodes and beneficial rhizosphere bacteria (actinomycetes) will be investigated to obtain a comprehensive picture on rhizosphere microbial communities. The project aims at getting detailed insight into the genetic and C allocation response of oak microcuttings to animal rhizosphere consumers, and the implications of these interactions for rhizosphere microorganisms.
