Description: Ziel dieser Studienerweiterung ist die Reduktion von Tierversuchen bei der Bestimmung der Zyto- und Gentoxizität von inhalierbaren Industriechemikalien (im Umfeld des Arbeits-, Verbraucher-, Gesundheits- und Umweltschutzes) durch eine standardisierte Alternativmethode. Die ersten Ergebnisse aus dem Vorhaben (FKZ.: 0313963B) zeigten, dass diese in der Prävalidierung befindliche Alternativmethode ein viel versprechendes Untersuchungskonzept darstellt. Andererseits wurde an den Ergebnissen auch deutlich, dass für eine abschließende, solide Beurteilung die Datenbasis erweitert werden muss. Die Studienerweiterung umfasst die Untersuchung von 5 weiteren Modellgasen (3 mit Toxizitätspotenzial und 2 Negativsubstanzen) und des Nachweises der grundsätzlichen Verwendbarkeit weiterer Endpunkte (Entzündungsmarker) in das zu validierende Prüfverfahren. Zur Erfassung der Intra- und Inter-Laborvariabilität werden die Prüfungen von 4 Laboren (aus den Bereichen des Arbeits-, Gesundheits-, Verbraucher-, Umweltschutzes und der Risikobewertung) durchgeführt und die erhobenen Daten statistisch zentral aufgearbeitet (BfR, AG Biometrie).

Title: Extended prevalidation study to test the toxic effects of inhalable substances (gases) after direct exposure of human lung cells at the air/liquid Interface - Part 3
Description: The acceptance of a more expanded application of a standardized, robust and predictive in vitro methodology for the toxicity evaluation after airborn exposures has improved significantly only in recent times (e.g. Draft proposals of OECD testing guideline 403, 436 and Guidance document 39; REACH regulation),This project aims to extend a current pre-validation study towards an in vitro methodology for the investigation of the toxic and genotoxic potential of inhalable substances by airborn exposure of the human lung cell line A-549 using the ALI technology ('ALI = air-liquid-interface'). Results of the first pre-validation Phase demonstrate a good intra- and inter-laboratory reproducibility. Against this background, the present follow-up project which mainly aims at the database extension, will be performed of the consortium (Fraunhofer-ITEM, Hannover; BAuA, Berlin; BfR-ZEBET, Berlin; UFZ-Leipzig). A total of four test gases with toxic potency and two test gases which serve as a negative control will be investigated. Based an the data material obtained and taking into account existing data from animal experiments (literature data), analyses will be performed regarding the relation between in vitro and in vivo studies and a prediction model (PM) will be developed. Before entering the international validation phase the enlargement of the database will therefore guarantee a secure assessment of this alternative method.
