Description: Das Projekt "Biogas: Abundanz und Vielfalt von Clostridien in landwirtschaftlichen Biogasanlagen unter besonderer Berücksichtigung von Clostridium botulinum" wird vom Umweltbundesamt gefördert und von HAWK Hochschule für angewandte Wissenschaft und Kunst - Hildesheim,Holzminden,Göttingen, Fachgebiet Nachhaltige Energie- und Umwelttechnik NEUTec durchgeführt. 1. The substrates, cattle manure, pig manure or maize silage contained different amounts of Clostridium cluster XIVa and cluster I, respectively . 2. There was no indication of an increase in the proportion of Clostridium cluster XIVa or cluster I during batch fermentation. Independent of the applied substrate, target gene copies ng-1 DNA were 106 for Bacteria and 105 for Archaea. In contrast, proportions of Clostridium cluster XIVa and cluster I were affected by the kind of substrate and its source (biogas plant). Gene copy numbers of Bacteria, Archaea and Clostridium cluster XIVa and cluster I of the batch experiment were reflected by those of the biogas plant. 3. Sequences obtained in preparation of high-throughput amplicon sequencing did not indicate the presence of C. botulinum. Introduction: Biogas is produced in reactors under anaerobic conditions by a consortium of microorganisms which commonly include bacteria of the genus Clostridium. Since the genus Clostridium also harbors some highly pathogenic members in its phylogenetic cluster I, e.g., Clostridium botulinum, there has been some concern that an unintended growth of such pathogens might occur during the fermentation process. A previous study revealed a selective effect of specific substrates used for biogas production on the bacterial and the clostridial community structures. However, the knowledge on the dynamics and on the presence of bacteria with lower abundance, particularly Clostridia is still very limited.
