Traditional Indonesian homegardens harbour often high crop diversity, which appears to be an important basis for a sustainable food-first strategy. Crop pollination by insects is a key ecosystem service but threatened by agricultural intensification and land conversion. Gaps in knowledge of actual benefits from pollination services limit effective management planning. Using an integrative and agronomic framework for the assessment of functional pollination services, we will conduct ecological experiments and surveys in Central Sulawesi, Indonesia. We propose to study pollination services and net revenues of the locally important crop species cucumber, carrot, and eggplant in traditional homegardens in a forest distance gradient, which is hypothesized to affect bee community structure and diversity. We will assess pollination services and interactions with environmental variables limiting fruit maturation, based on pollination experiments in a split-plot design of the following factors: drought, nutrient deficiency, weed pressure, and herbivory. The overall goal of this project is the development of 'biodiversity-friendly' land-use management, balancing human and ecological needs for local smallholders.

Rain-cracking limits the production of many soft and fleshy fruit including sweet cherries world wide. Cracking is thought to result from increased water uptake through surface and pedicel. Water uptake increases fruit volume, and hence, turgor of cells (Pcell) and the pressure inside the fruit (Pfruit) and subjects the skin to tangential stress and hence, strain. When the strain exceeds the limits of extensibility the fruit cracks. This hypothesis is referred to as the Pfruit driven strain cracking. Based on this hypothesis cracking is related to two independent groups of factors: (1) water transport characteristics and (2) the intrinsic cracking susceptibility of the fruit defined as the amount of cracking per unit water uptake. The intrinsic cracking susceptibility thus reflects the mechanical constitution of the fruit. Most studies focussed on water transport through the fruit surface (factors 1), but only little information is available on the mechanical constitution (i.e., Pfruit and Pcell, tensile properties such as fracture strain, fracture pressure and modulus of elasticity of the exocarp; factors 2). The few published estimates of Pfruit in sweet cherry are all obtained indirectly (calculated from fruit water potential and osmotic potentials of juice extracts) and unrealistically high. They exceed those measured by pressure probe techniques in mature grape berry by several orders of magnitude. The objective of the proposed project is to test the hypothesis of the Pfruit driven strain cracking. Initially we will focus on establishing systems of widely differing intrinsic cracking susceptibility by varying species (sweet and sour cherry, Ribes and Vaccinium berries, plum, tomato), genotype (within sweet cherry), stage of development and temperature. These systems will then be used for testing the hypothesis of Pfruit driven strain cracking. We will quantify Pfruit und Pcell by pressure probe techniques and compression tests and the mechanical properties of the exocarp using biaxial tensile tests. When the presence of high Pfruit and Pcell is confirmed by direct measurements, subsequent studies will focus on the mode of failure of the exocarp (fracture along vs. across cell walls) and the relationship between failure thresholds and morphometric characteristics of the exocarp. However, when Pfruit und Pcell are low, the hypothesis of Pfruit driven strain cracking must be rejected and the mechanistic basis for low pressures (presence of apoplastic solutes) clarified on a temporal (in the course of development) and a spatial scale (exocarp vs. mesocarp). We focus on sweet cherry, because detailed information on this species and experience in extending the short harvest period is available. Where appropriate, other cracking susceptible species (sour cherry, plum, Vaccinium, Ribes, tomato) will be included to further extend the experimental period and to maximize the range in intrinsic cracking susceptibility.
Steroid hormones are essential in orchestrating oocyte maturation, i.e. estrogens of follicular origin support the development of the female gamete and fertilization. In this project the concentration of free and conjugated estrogens during follicular development will be analysed and compared to local concentrations in the developing follicle. Cattle are suitable animal models because of the accessibility and suitability for frequent examination and sampling. Furthermore, it has been useful for understanding several features of human reproduction including follicular dynamics, the fate of the emerging follicles is orchestrated mainly by gonadotropins and steroid hormones in a similar manner. Ovarian SULT1E1 participates locally in the regulation of follicular estrogen activity. The ESTcatalysed down-regulation of estrogen activity enables normal ovulation. Conversely, sulfoconjugated estrogens may also be precursors of the production of free estrogens depending on estrogen sulfatase (StS) acitivity. In mammals, follicular luteinisation/ovulation is triggered by a surge in LH and is characterised by numerous physical and biochemical changes, including the decreased production of estradiol (E2). This loss in E2 biosynthetic capacity has been explained by a marked decrease in the expression of key steroidogenic enzymes involved in the follicular production of active estrogens. However, little is known about the regulation of enzymes/proteins responsible for the inactivation and elimination of estrogens, as mediated for example by EST during this period.
bifa hat ein Vorhaben für die G8- Staaten bearbeitet, in dem die Entwicklungen in Deutschland innerhalb der neun Handlungsfelder ( Actions ) des Kobe 3R Action Plan dargestellt werden. Mit der 3R-Initiative beabsichtigen die G8-Staaten seit 2004 eine bessere Verankerung der Nachhaltigkeit im Umgang mit Rohstoffen durch die stärkere Förderung der drei Prinzipien Reduce, Reuse, Recycle , abgekürzt 3R , in den nationalen Abfallwirtschaftspolitiken. Im Rahmen der Beauftragung untersuchte bifa, welche Punkte aus dem Kobe 3R Action Plan bereits hinreichend durch bestehende Entwicklungen bzw. ergriffene Maßnahmen abgedeckt sind, bei welchen Aktionen noch Lücken bestehen und wie diese Lücken gefüllt werden können. Legt man die drei Zielsetzungen des Kobe 3R Action Plan und die ihnen zugeordneten Handlungsfelder als Prüfraster über die deutsche Abfallwirtschaftspolitik, lässt sich ein sehr hoher Erfüllungsgrad feststellen. Ein erheblicher Teil der vorgeschlagenen Handlungsoptionen war in Deutschland bereits vor 2008 durch konkrete Maßnahmen umgesetzt worden. Für einen anderen Teil wiederum lässt sich der Ursprung, z. B. in Form eines ersten Gesetzentwurfs, auf die Zeit vor 2008 zurückdatieren, die Umsetzung durch die Veröffentlichung im Bundesgesetzblatt aber fand 2008-2011 statt. Einige Regelungen setzen Richtlinien oder Verordnungen der EU, die ihrerseits zum Teil auf Bestrebungen Deutschlands hin zustande kamen, in nationales Recht um. Mit dem in einer fortgeschrittenen Version vorliegenden Entwurf eines novellierten Kreislaufwirtschaftsgesetzes vollzieht Deutschland einen weiteren wichtigen Schritt hin zu einer Abfallwirtschaft, deren Markenzeichen insbesondere eine hohe Ressourceneffizienz ist. Dennoch verbleiben Optimierungspotenziale, zu deren Ausschöpfung bifa Vorschläge für das Bundesumweltministerium erarbeitet hat. Im Zuge des Projekts analysierte bifa u. a. die Importe und Exporte notifizierungspflichtiger Abfälle. Der Saldo hat sich den bifa-Analysen zufolge seit 1998 umgekehrt: Wurden 1998 noch etwa doppelt so viel notifizierungspflichtige Abfälle exportiert wie importiert, hat sich der Import seitdem vervierfacht und die Exporte sind sogar leicht gesunken. Ein wichtiger Grund ist die Verfügbarkeit von Behandlungs- und Verwertungskapazitäten von hoher Leistungsfähigkeit in Deutschland. Die Schadstoffentfrachtung von Abfällen aus Ländern mit einer wenig entwickelten Entsorgungsinfrastruktur führt jedoch innerhalb der deutschen Öffentlichkeit immer wieder zu Kontroversen. The biotrophic fungus Ustilago maydis infects corn and induces the formation of tumors. In order for the fungus to proliferate in the infected tissue, U. maydis has to redirect the metabolism of the host to the site of infection. We wish to elucidate how this is accomplished. To this end we will perform transcript profiling during the time course of infection for both, the fungus and the maize plant. This will be complemented by metabolome analysis of different tissues during infection as well as by apoplastic fluid analysis. The goals will be to identify the carbon sources taken up by the fungus during biotrophic growth, to identify the transporters required for uptake, determine their specificity and elucidate how these carbon sources are provided by the plant. Fungal mutants affected in discrete stages of pathogenic development will be included in these studies. Likely candidate genes for carbon uptake/supply as well as for redirecting host metabolism will be functionally characterized by generating knockouts in the fungus and by isolating plants carrying mutations in respective genes or by generating transgenic plants expressing RNAi constructs.
Boron (B) is an essential microelement for plants. Despite the use of modern fertilization methods, B deficiency still causes losses in agricultural plant production. Even though many positive effects of B on plant growth and physiology have been reported, a large majority of B functions and the regulatory mechanisms controlling the B nutritional status remain unknown. The main objective of this project is to elucidate how the greatly B deficiency-sensitive Brassica crop plants process and regulate their B status during vegetative and reproductive growth. In this context, the project aims at identifying the mode of action of B in mechanisms regulating the B status itself and uncovering those mechanisms contributing to B efficiency in different genotypes. Plant species subjected to investigation will be the agronomically important oilseed and vegetable plant Brassica napus (rapeseed) and its close relative the genetic and molecular model plant Arabidopsis thaliana. Questions addressed within the scope of this project should lead to a detailed understanding of mechanisms controlling B uptake and allocation from the level of the whole plant down to the cellular level. B transport routes and rates will be determined in sink- and source tissues and in developmental periods with a particularly high B demand. A special focus will be on the identification of B transport bottlenecks and the analysis of B deficiency-sensitive transport processes to and within the highly B-demanding reproductive organs. Recent studies in Arabidopsis suggest that Nodulin26-like Intrinsic Proteins (NIPs), which belong to the aquaporin channel protein family, are essential for plant B uptake and distribution. The systematic focus on the molecular and physiological characterization of B. napus NIPs will clarify their role in B transport and will identify novel NIP-associated mechanisms playing key roles in the B response network.To further resolve the mostly unknown impact of the B nutritional status on gene regulation and metabolism, a transcript and metabolite profile of B-sufficient and B-deficient rapeseed plants will be generated. Additionally, an Arabidopsis transcription factor knockout collection (greater 300 lines) will be screened for abnormalities in responses to the B nutritional status. This will identify yet unknown B-responsive genes (transcription factors and their targets) and gene products (enzymes or metabolite variations) playing key roles in signalling pathways and mechanisms regulating the B homeostasis. Boron (in form of boric acid) and arsenite (As) share in all likelihood the same NIP-mediated transport pathways. To assess the consequences of this dual transport pathway the so far unstudied impact of the plants B nutritional status on the accumulation and distribution of As will be investigated in B. napus. Moreover, the current dimension of the As contamination of Brassica-based food products, to which consumers are exposed to, will be analyzed. usw.
Cydia pomonella granulovirus (CpGV, Baculoviridae) is one of the most important agents for the control of codling moth (CM, Cydia pomonella, L.) in both biological and integrated pest management. The rapid emergence of resistance against CpGV-M, which was observed in about 40 European CM field populations from 2003 on, could be traced back to a single, dominant, sex-linked gene. Since then, resistance management has been based on mixtures of new CpGV isolates (CpGV-I12, -S), which are able to overcome this resistance. Recently, resistance even to these novel isolates was observed in CM field populations. This resistance does not follow the described dominant, sex-linked inheritance trait. At the same time, another isolate CpGV-V15 was identified showing high virulence against these resistant populations. To elucidate this novel resistance mechanism and to identify the resistance gene(s) involved, we propose a comprehensive analysis of this resistance on the cellular and genomic level of codling moth. Because of the lack of previous knowledge of the molecular mechanisms of virus resistance in insects, several different and complementary approaches will be pursued. This study will not only give an in-depth insight into the genetic possibilities for development of baculovirus resistance in CM field populations and how the virus overcomes it, but can also serve as an important model for other baculovirus-host interaction systems.
