Adult Littorina littorea were collected from the intertidal zone of Helgoland (North Sea, Germany; 54°11′ N, 7°53′ E) during November 2023 and June 2024 to investigate physiological and behavioral responses to acute warming. After collection, the organisms were transported to the Alfred Wegener Institute in Bremerhaven, Germany, and acclimated for two weeks to season-specific temperatures representative of winter (10 °C) and summer (16 °C) in aquarium systems containing filtered, recirculating, and continuously aerated seawater at a salinity of 33. Animals were fed Fucus vesiculosus ad libitum throughout the acclimation period. Following acclimation, individuals were exposed to a continuous warming ramp from the respective seasonal acclimation temperature to 34 °C at a rate of 1.5 °C h⁻¹. Samples were collected at predefined temperature intervals between 10 and 34 °C for winter-acclimated animals and between 16 and 34 °C for summer-acclimated animals.The dataset contains behavioral, metabolomic, and biochemical measurements obtained from snails at the respective experimental temperatures. Behavioral observations include the time required for antenna extension and the initiation of locomotion. Metabolomic data were obtained from whole-body soft-tissue extracts using proton nuclear magnetic resonance spectroscopy using the ultra-shielded vertical 9.4 T NMR spectrometer (Advance III HD 400 WB, Bruker-BioSpin GmbH, Germany), with a 1.7 mm triple tuned 1H-13C-15N NMR probe and a Carr-Purcell-Meiboom-Gill sequence (Bruker protocol cpmgpr1d, TOPSPIN 3.5, Bruker GmbH, Germany) with water suppression. Biochemical measurements comprise heat shock protein 70 abundance and catalase activity determined from whole-body tissue homogenates. The data were collected to characterize temperature-dependent physiological and behavioral responses of L. littorea across seasons and to identify thermal thresholds during acute warming exposure.
Organisms in intertidal zones experience fluctuations in environmental stressors such as hypoxia and temperature. These stressors and their fluctuations often appear in combination. Combination of stressors can have different effects compared to single stressors. In this study, we investigate the physiological effects of intermittent hypoxia in combination with different temperature regimes on the Pacific Oyster Crassostrea (Magallana) gigas. The oysters were exposed to hypoxic cycles (12h hypoxia by emersion/12h submersion) at normal (15°C), elevated (30°C) or fluctuating (15°C submersion/30°C emersion) temperature for 10 days. After the last submersion phase, the gills and digestive gland were sampled. We measured markers for bioenergetics and redox-balance in the gills and digestive gland using colorimetric methods as well as a set of metabolites (predominantly amino acids, osmolytes, anaerobic end products and energetic metabolites) in the gills using LC-MS/MS. Oysters kept submerged for up to 10 days were used as controls.
Objective: Engineered Nanoparticles (ENP) are increasingly produced for use in a wide range of industrial and consumer products. Yet it is known that exposure to some types of particles can cause severe health effects. Therefore it is essential to ascertain whether exposure to ENP can lead to possible health risks for workers and consumers. We have formed a consortium of well-known scientists from European Universities and Research Institutes, with over 100 publications in the field of Nanotoxicology. Our aim is to develop an approach for the Risk Assessment of ENP (ENPRA). Our objectives are: (i) to obtain a bank of commercial ENP with contrasting physico-chemical characteristics and measure them; (ii) to investigate the toxic effects of ENP on 5 (pulmonary, hepatic, renal, cardiovascular and developmental) target systems and 5 endpoints (oxidative stress, inflammation; immuno-toxicity; fibrogenecity; genotoxicity) using in vitro animal/human models; (iii) to validate the in vitro findings with a small set of carefully chosen in vivo animal experiments; (iv) to construct mathematical models to extrapolate the exposure-dose-response relationship from in vitro to in vivo and to humans; (v) to use QSAR like models to identify the key ENP characteristics driving the adverse effects; (vi) to implement a risk assessment of ENP using the Weight-of-Evidence approach; (vii) to disseminate our findings to potential stakeholders. To harmonise the research activities between our EU group and the US, we have established links with scientists from US Universities (Duke, Rochester) and Government Agencies (NIH/NIEHS, NIOSH and EPA) with on-going research in Nanotoxicology. Our objectives here are: (vii) to share information and agree on experimental protocols; (viii) to avoid duplication of work; (ix) to further validate the findings of this proposed study.
In vielen Organismen erfolgt die Biotransformation der cyanobakteriellen Microcystine über die Konjugation an Glutathion. Während der weiteren Biotransformationsprozesse, in welchen das Glutathionkonjugat weiter abgebaut wird, kann es zur Entstehung reaktiver Sauerstoffspezies kommen. Diese aktivieren ein antioxidatives Abwehrsystem, und es werden Enzyme wie die Superoxiddismutase, Katalase, Glutathionperoxidase in ihrer Aktivität gesteigert. Auch der Ascorbat-Glutathion Kreislauf wird beeinflusst. Während der Exposition von Ceratophyllum demersum konnte die Bildung eines Microcystin-LR Glutathionkonjugates nachgewiesen werden, ebenso die Aktivierung von löslichen und mikrosomalen Glutathion STransferasen, die Beeinflussung des Glutathionpools, sowie, die Aktivierung verschiedenster antioxidativer Enzyme.
This project aims to identify suitable early toxicology indicator for nanoparticle and elucidation of major parameters governing nanoparticles cyto-toxicology with a focus on oxide nanoparticles.
Dieser Überblick beginnt mit der Beschreibung quantitativer Verhältnisse zwischen toter (Detritus, Huminstoffe) und lebender organischer Biomasse in verschiedenen Ökosystemen. In den meisten Binnengewässern stammt die größte HS-Menge aus der terrestrischen Umgebung und sind Lignin- Abbauprodukte. Trotz der großen Mengen von Huminstoffen (HS) in den Ökosystemen ist das Wissen über die ökologische Bedeutung dieser Stoffe gering und voll von alten, immer wiederholten Paradigmen. So hält man HS für inert oder zumindest refraktär und zu groß, um von aquatischen Organismen aufgenommen zu werden. In diesem Überblick zeige ich, dass HS aufgenommen werden und danach sowohl indirekt als auch direkt mit aquatischen Organismen interagieren und so Biozönosen strukturieren können. Relativ gut untersucht ist inzwischen die alimentierende Funktion von allochthonen HS, die nach Belichtung kurzkettige Fettsäuren freisetzen, die als Substrat für mikrobielles Wachstum dienen. Dies ist ein indirekter Effekt. Mikroben ihrerseits sind Nahrung für mixotrophe Algen und heterotrophes Zooplankton. Deshalb sind nicht-eutrophierte Gewässer netto-heterotroph: die Atmung übersteigt die Photosynthese. Weiterhin zeigen Modellkalkulationen, dass nur eine sehr geringe Menge der terrestrischen Primärproduktion ausreicht, die Netto-Heterotrophie in Gewässern sicherzustellen. Jüngste Arbeiten zeigen auch, dass durch die stöchiometrische Zusammensetzung der Bakterien die maximale Planktonproduktion geringer ist, wenn Bakterien und nicht Algen der Beginn der Nahrungskette sind. Ebenfalls kürzlich wurden verschiedene direkte Effekte von HS auf aquatische Organismen gefunden. Hierzu gehören: Induktion von Chaperonen (stress shock proteins), Modulation der Biotransformationsenzyme, Modulation (überwiegend Hemmung) der photosynthetischen Sauerstofffreisetzung durch aquatische Pflanzen, Entwicklung eines internen oxidativen Stresses, Modulation der Nachkommenzahl im Nematoden Caenorhabditis elegans, Feminisierung bei Fischen und Amphibien, Interferenz innerhalb des Schilddrüsensystems und Wirkung als Lockstoff. Bei diesen Effekten sind wir noch in der Phase, die verschiedenen physiologischen, biochemischen und molekular- biologischen Effekte zu identifizieren. Deshalb sind die ökologische und ökophysiologische Signifikanz noch relativ unklar. Gleichwohl beeinflussen die HS sowohl Individuen als auch Gemeinschaften und ganz Ökosysteme, ein Einfluss, der mit dem von Nährstoffen vergleichbar ist.
