Die pflanzliche Photosynthese ist der zentrale Mechanismus in der Natur, Sonnenenergie in chemische Energie umzuwandeln. Darum wird die mikrobielle Photokatalyse als effiziente und nachhaltige Alternative für derzeitige chemische Prozesse angesehen. Während herkömmliche Photobioreaktoren mit Suspensionkulturen durch geringe Zelldichten begrenzt sind, könnten Biofilme als selbstimmobilisierte Mikroben mit hohe Zelldichte dieses Problem überwinden. Jedoch stellen hohe Zelldichte und Stabilität gegenüber variierenden Umweltbedingungen, wie Strahlung und Temperatur, eine Herausforderung für stabile katalytische Produktionsprozesse dar. Das Projekt zielt darauf, das Prinzip von Pflanzenblättern mit Selbstregulation und stabiler Unterhaltung der Photosynthesemaschinerie zu adaptieren. Mit Hilfe gentechnisch modifizierter phototropher Biofilme, die in einem porösen Zellkulturträger mit hoher Zelldichte wachsen, wobei dieses Material mit einer responsiven Hydrogelschicht an der Gas-Flüssig-Grenzfläche ausgestattet ist, soll eine stabile biokatalytische Aktivität für die chemische Produktion realisiert werden. Die pH- oder Temperatur (T)-responsiven Hydrogelschichten werden auf den Aktivitätszustand der mikrobiellen Konsortien innerhalb des porösen Materials reagieren und so eine Selbstkontrolle der katalytischen Aktivität durch Steuerung der Gaspermeation ermöglichen, ähnlich wie Pflanzenblattoberflächen. Das Konzept eines "mikrobiellen Blatts" wird umgesetzt auf Basis unserer Expertise zu mikrobiellen Konsortien für die chemische Produktion (z.B. Synechocystis, Pseudomonas) und der Entwicklung von Zellkulturträgern, einschließlich der Synthese von pH- und T-responsiven Hydrogelen, um eine selbstkontrollierte Aktivität eines lebenden katalytischen Materials (LCM) zu zeigen. In unserer Fallstudie werden wir die Leistungsfähigkeit von mikrobiellen Biofilmen in porösen Polymermaterialien für die katalytische Umwandlung von Cyclohexan in ?-Caprolacton oder Adipinsäure als Vorstufen der industriellen Polymersynthese untersuchen. Es soll gezeigt werden, dass Schwach- und Starklichtzustände, ähnlich dem Tag-Nacht-Zyklus, die Mikroumgebung der LCM so beeinflussen, dass dies zu gequollenen und kollabierten Zuständen der pH-responsiven Hydrogelschicht führt. Diese responsive Hydrogelschicht wird durch elektronenstrahlinitiierte Polymerisation von N-Isopropylamid- und Acrylsäuremonomeren mit einer Anpassung der Phasenübergangsbereichen synthetisiert. Die unterschiedlichen Quellungszustände und physikochemischen Eigenschaften der Hydrogelschicht, die durch die Aktivität der Biofilme ausgelöst werden, steuern den Zufluss von Cyclohexan für die katalytische Reaktion der adaptiven LCM, wodurch eine optimale Leistungsfähigkeit bei variierenden Lichtverhältnissen ermöglicht wird. In Zukunft werden unsere Erkenntnisse das Potential des Mikrobiellen-Blatt-Konzepts ausschöpfen lassen und selbststabilisierte biotechnologische Prozesse mit breiter Anwendbarkeit ermöglichen.
Maeuse inhalieren 0,5-3 tg lang Campher oder Cyclohexan. Es wird die Zunahme an cyt. P-450, der nadph-Cyt. p-450-Reduktase sowie der Aethylumbelliferon-Desalkylase bestimmt. Es soll vor allem die untere Grenze der Wirksamkeit der Induktoren und ihre Konzentration in Blut und Leber bestimmt werden. Weiter soll der Stoffwechsel der Induktoren in Abhaengigkeit von der Induktionszeit in vivo und in vitro untersucht werden. Campher wird in der Medizin als Einreibungsmittel benutzt und kommt in Konservierungs- und Desinfektionsmitteln sowie in Mottenkugeln vor. Cyclohexan ist Bestandteil bestimmter Benzinarten.
Bei einem 3-methylcholanthrenempfindlichen Maeusestamm (C57BL/6N) soll geprueft werden, ob chronische orale Verabreichung von Aethanol in der Leber sowohl die mikrosomale Aethanoloxidation als auch die Dehydrierung von Aethanol durch die Alkohol-Dehydrogenase des Lebercytosols induziert. Die induzierende Wirkung des Aethanols soll mit derjenigen von D,L-Campher und Cyclohexan verglichen werden. Es soll versucht werden, die Induktionen durch Induktionshemmer zu verhindern. Inzwischen ist nachgewiesen worden: In den Lebermikrosomen induziert orale chronische Verabreichung von Aethanol MEOS bei maennlichen C57-Maeusen in 4 Monaten, um den Faktor 2-3. Die Induktion ist bereits nach 14 Tagen nachweisbar und durchlaeuft ein Minimum nach 1,5 Monaten. Alkohol-Dehydrogenase wird im Cytosol anscheinend nicht induziert, wohl aber andere Proteine, denen moeglicherweise Acetaldehyd-Dehydrogenaseaktivitaet zukommt. Diese induzierbaren Nicht-Haem-Proteine erscheinen auch vermehrt nach Vorbehandlung der Maeuse mit anderen Induktoren. Es soll 1982 geprueft werden, wie weit die Dehydrierung von Acetaldehyd durch Aethanol induziert werden kann, und ob andere Induktoren diese nicht wirksamer induzieren.
technologyComment of adipic acid production (RER, RoW): This dataset models the production of adipic acid by nitric acid oxidation of a mixture of cyclohexanol and cyclohexanone, which is obtained by oxidation of cyclohexane. Abatement of N2O emissions is assumed to reduce the emissions by 80%. Essentially all production of adipic acid is derived from the nitric acid oxidation of a mixture of cyclohexanone – cyclohexanol (KA mixture). The reactor, controlled at 60 – 80 °C and 0.1 – 0.4 MPa, is charged with the recycled nitric acid stream, the KA feed material, and makeup acid containing 50 – 60 % nitric acid and copper – vanadium catalyst. The reaction is very exothermic (6.280 MJ/kg). Adipic acid is obtained with a yield greater than 90%. Nitrogen oxides, carbon dioxide, and some lower dicarboxylic acids are the major by-products, as well as oxidation products arising from impurities in the KA intermediate. The nitric acid oxidation step produces three major waste streams: an off-gas containing oxides of nitrogen and CO2, water containing traces of nitric acid and organics from the water removal column; and a dibasic acid purge stream containing adipic, glutaric and succinic acids. technologyComment of cyclohexane production (RER, RoW): Over 90 % of all cyclohexane is produced commercially by hydrogenation of benzene. A small amount is produced by superfractionation of the naphtha fraction from crude oil. Naturally occurring cyclohexane can be supplemented by fractionating methylcyclopentane from naphtha and isomerizing it to cyclohexane. Hydrogenation of benzene: Benzene can be hydrogenated catalytically to cyclohexane in either the liquid or the vapor phase in the presence of hydrogen. Several cyclohexane processes, which use nickel, platinum, or palladium as the catalyst, have been developed. Usually, the catalyst is supported, e.g., on alumina, but at least one commercial process utilizes Raney nickel. Hydrogenation proceeds readily and is highly exothermic (Δ H500K = – 216.37 kJ/mol). From an equilibrium standpoint, the reaction temperature should not exceed 300 °C. Above this, the equilibrium begins to shift in favor of benzene so that high-purity cyclohexane cannot be produced. As a result of these thermodynamic considerations, temperature control of the reaction is critical to obtaining essentially complete conversion of benzene to cyclohexane. Temperature control requires economic and efficient heat removal. This has been addressed in a number of ways by commercial processes. The earlier vapor-phase processes used multistage reactors with recycle of cyclohexane as a diluent to provide a heat sink, staged injection of benzene feed between reactors, and interstage steam generators to absorb the exothermic heat of hydrogenation. In the 1970s processes have been developed that use only one reactor or a combination of a liquid-and a vaporphase reactor. The objectives of the later processes were to reduce capital cost and improve energy utilization. However, all of the commercial processes have comparably low capital cost and good energy efficiency. In the vapor-phase process with multistage reactors in series, the benzene feed is divided and fed to each of the first two reactors. Recycled cyclohexane is introduced to the first reactor along with hydrogen. The recycled cyclohexane enables higher conversion in the reactors by absorbing part of the heat of hydrogenation. Steam generators between the reactors remove the heat of hydrogenation. The outlet temperature of the last reactor is controlled to achieve essentially 100 % conversion of benzene to cyclohexane. The effluent from the last reactor is cooled, and the vapor and liquid are separated. Part of the hydrogen-rich vapor is recycled to the first reactor, and the rest is purged to fuel gas or hydrogen recovery facilities. The liquid from the separator goes to a stabilizer where the overhead gas is sent to fuel gas; the remaining material is cyclohexane product, part of which is recycled to the first reactor. In the process with liquid- and vapor-phase reactors, benzene and hydrogen are fed to the liquid-phase reactor, which contains a slurry of finely divided Raney nickel. Temperature is maintained at 180 – 190 °C by pumping the slurry through a steam generator and by vaporization in the reactor. Roughly 95 % of the benzene is converted in this reactor. The vapor is fed to a fixed-bed reactor where the conversion of benzene is completed. The effluent from the fixed-bed reactor is processed as described previously for the vapor-phase process. Benzene hydrogenation is done typically at 20 – 30 MPa. The maximum reactor temperature is limited to ca. 300 °C so that a typical specification of < 500 mg/kg benzene and < 200 mg/kg methylcyclopentane in the product can be achieved. This is necessary because of the thermodynamic equilibrium between cyclohexane – benzene and cyclohexane – methylcyclopentane. Actually, equilibrium strongly favors methylcyclopentane, but the isomerization reaction is slow enough with the catalysts employed to avoid a problem if the temperature is controlled. The hydrogen content of the makeup hydrogen has no effect on product purity but it does determine the makeup, recycle, and purge gas rates. Streams with as low as 65 vol % hydrogen can be used. Carbon monoxide and sulfur compounds are catalyst deactivators. Both can be present in the hydrogen from catalytic naphtha reformers or ethylene units, which are typical sources of makeup hydrogen. Therefore, the hydrogen-containing stream is usually passed through a methanator to convert carbon monoxide to methane and water. Prior to methanation, hydrogen-containing gas can be scrubbed with caustic to remove sulfur compounds. Commercial benzene contains less than 1 mg/kg sulfur. In some cases, the recycle gas is also scrubbed with caustic to prevent buildup of hydrogen sulfide from the small amount of sulfur in the benzene. With properly treated hydrogen and specification benzene, a catalyst life in excess of three years can be achieved easily in fixed-bed reactors that use noble-metal catalysts supported on a base. Aufgabenbeschreibung: Der Mensch atmet pro Tag 10 bis 20 m3 Luft ein, was einer Masse von 12 bis 24 kg Luft pro Tag entspricht. Davon entfällt etwa 90 % auf die Innenraumluft. Damit kann kontaminierte Innenraumluft zu einer relevanten Aufnahme von Chemikalien führen. Es ist deswegen äußerst wichtig, die Vorkehrungen zu treffen, die eine gute Innenraumluftqualität sicherstellen. Für diesen Zweck setzt der Ausschuss für Innenraumrichtwerte (AIR) bundeseinheitliche, gesundheitsbezogene Richtwerte für die Innenraumluft fest. Der AIR hat bereits über 100 Stoffe in der Innenraumluft bewertet und für eine Vielzahl von Stoffen Richtwerte abgeleitet, die als Maßstab für die Bewertung der Innenraumluftqualität öffentlicher und privater Gebäude in Deutschland angewandt werden. Wegen der Entwicklung der Bautechnik, kultureller Veränderungen und neuer wissenschaftlicher Erkenntnisse werden stets neue Stoffe in der Innenraumluft identifiziert. Die Liste der Stoffe für die eine toxikologische Bewertung dringend notwendig ist, wurde durch den Ausschuss für Innenraumrichtwerte im Jahr 2016 aktualisiert. Gemäß der Angaben der beteiligten Länder besteht ein akuter Bedarf nach Richtwerten für Pentan, Methylpentan, Hexan, Cyclohexan und Summenrichtwerten für C4-C8 Alkane. Ziel des Projekts ist die Bereitstellung eines geeigneten Stoffdossiers als Bewertungsgrundlage für Ableitung der Richtwerte durch den AIR.

The feasibility of an in-situ contaminant phase extraction of highly-chlorinated hydrocarbons below the dump site of production residues using co-solvent flooding (ternary solvent-mixtures) was investigated under contract of an international chemical company. Both cyclohexane and diethyl ether were selected as co-solvents because of their high partitioning capacity into the contaminant mixture and their ability to transform the contaminant mixture (DNAPL) into a single-phase LNAPL. Additionally, the selected co-solvents were able to dissolve the precipitated tar oil phase. The water-miscible solvents were acetone and 2-propanol. Four different ternary phase diagrams (TPD) for the solvent-co-solvent-water system were determined in order to define the optimal co-solvent flushing (CSF)-mixture. No significant differences were observed between the selected CSF-cocktails for a composition of 90/10 %v/v. Therefore, after considering the costs and safety and ecological aspects, the solvent 2-propanol and the co-solvent cyclohexane were selected. Solids and/or highly viscous tar oil precipitated during the determination of the ETPDs. The hydraulic control of the remediation process may be compromised by a clogging of the aquifer during CSF. The cost of the chemicals and the recycling of the CSF-cocktail were estimated after the recycling process was designed. Due to the physical and chemical properties of the chemicals, a thermal recycling of the CSF-cocktail by vacuum evaporation was proposed. The costs for the recycling process and the overall costs for the CSF-cocktail needed for the removal of the contaminants were estimated based on the results of previous research work. The costs are roughly estimated to be 800 - 900 €/m3 treated soil for the chemicals and the recycling of the CSF-cocktail. A more accurate cost estimate must be based on further lab experiments to determine the mass fluxes, the effectiveness and the functionality of the process.
Im Rahmen des Projekts werden experimentelle und theoretische Untersuchungen zur Kopplung der gleichgewichtslimitierten Dehydrierung von Cyclohexanol mit der exothermen Hydrierung von Cyclohexen in einem katalytischen Membranreaktor durchgefuehrt. In dem am Institut konzipierten Laborreaktor werden die im Membranreaktor simultan ablaufenden Prozesse zuerst separat voneinander charakterisiert. Dieses sind u a die chemische Reaktion und der Stofftransport durch Membranen. Anschliessend werden beide Prozesse gekoppelt. Die Untersuchungen zeigen, dass die Reaktions- und Permeationskinetik hinreichend gut mit entsprechenden Modellen beschrieben werden koennen. Weiterhin hat die Kopplung beider Prozesse ergeben, dass in Membranreaktoren hoehere Umsaetze erzielt werden, als in konventionellen Reaktoren.
Planung der Modellanlage zur Reinigung der Abwaesser der Cyclohexanoxydation weitgehend abgeschlossen. Auf den Bau der urspruenglich geplanten kleinen Pilotanlage wird verzichtet, stattdessen soll sofort eine Modellanlage im kleinen Betriebsmassstab erstellt werden. Schwerpunkt: Untersuchungen zur Anwendbarkeit der Nassoxidation bei den verschiedenen chemischen Stoffklassen bzw. Industrieabwaessern mit unterschiedlichen organischen Belastungen.
